NEW BRUNSWICK (dpa-AFX) - Johnson & Johnson (JNJ) reported encouraging Phase 1 clinical data for JNJ-6143, an investigational oral therapy for advanced prostate cancer, highlighting significant anti-tumor activity in patients who had previously undergone multiple lines of treatment.
JNJ-6143, formerly known as HLD-0915, is a once-daily oral Regulated Induced Proximity Targeting Chimera (RIPTAC) small molecule designed to selectively target cancer cells. The study evaluated the drug in patients with advanced prostate cancer who had exhausted several prior treatment options, an area where unmet medical need remains high.
Among 16 patients with measurable disease, 75% achieved a confirmed complete or partial response based on Response Evaluation Criteria in Solid Tumors (RECIST), demonstrating substantial tumor shrinkage. The response rate underscores the therapy's potential to deliver meaningful clinical benefit in a heavily pretreated patient population.
The treatment also generated notable reductions in prostate-specific antigen (PSA), a key biomarker used to monitor prostate cancer. At the 50 mg dose level, 77% of patients experienced a confirmed PSA decline of at least 50%, while 48% achieved PSA reductions of at least 90%, suggesting robust biological activity against the disease.
In addition to its efficacy signals, JNJ-6143 demonstrated a favorable safety profile. Most treatment-related adverse events were reported as mild to moderate, supporting continued clinical development of the therapy.
JNJ-6143 is an investigational, once-daily molecule that uses the novel RIPTAC platform. Its targeted dual-protein lock brings together BRD4, an essential protein for cell survival, and the tumor-selective protein, androgen receptor (AR), which is highly expressed in prostate cancer cells, and is designed to disrupt cancer cell function and trigger cancer cell death.
JNJ-6143 is being evaluated in Phase 1 and Phase 2 studies in patients with metastatic prostate cancer. It is the first investigational compound from the RIPTAC platform to advance into clinical studies.
In December 2025, Johnson & Johnson completed its acquisition of Halda Therapeutics, adding JNJ-6143 and the RIPTAC platform to its oncology portfolio.
JNJ closed Friday's regular trading at $263.81 down $3.80 or 1.42%.
